چکیده

The specificity of antibodies for different antigens appears to result from differences in the amino acid sequences of both the heavy and the light polypeptide chains of which antibody molecules are composed. -4 Although specific antibodies are too heterogeneous for complete amino acid sequence analysis, homogeneous iminunoglobulins may be obtained for this purpose from various plasma cell tumors. Bence-Jones proteins have been shown to be light polypeptide chains5' 6 and data have accumulated on the amino acid sequences of various samples of both humanl-9 and murine origin.'0'" These amino acid sequence analyses show that light polypeptide chains of type K (K chains) are composed of 212-214 amino acid residues.'2 From residue 1

Helicobacter pylori (H. pylori) is the causative agent in development of gastroduode-nal diseases, such as chronic atrophic gastritis, peptic ulcers, mucosa associated lym-phoid tissue (MALT) lymphoma, and gastric cancer. H. pylori has been associated with inflammation in cardia, showing the fact that infection with this bacterium could also be a risk factor for gastric cardia cancer. Gastric c...

Secondary mechanisms of diversification in the human antibody repertoire

V(D)J recombination and somatic hypermutation (SHM) are the primary mechanisms for diversification of the human antibody repertoire. These mechanisms allow for rapid humoral immune responses to a wide range of pathogenic challenges. V(D)J recombination efficiently generate a virtually limitless diversity through random recombination of variable (V), diversity (D), and joining (J) genes with div...

Evolution of Antibody Structure during the Immune Response

How an animal develops a state of antigen-specific, humoral immunity during the course of a primary immune response is arguably one of the most long-standing questions in the field of immunology. The clonal selection hypothesis (1, 2) explains this phenomenon by assuming that immunity is elaborated by an antigen-selective mechanism that draws from a large preexisting repertoire of B lymphocytes...

Construction of the Recombinant Plasmid Expressing AID under the Control of Temperature-sensitive Promoter of Bacteriophage Lambda

Background and purpose: Activation-induced cytidine deaminase (AID) is a B-cell specific enzyme responsible for somatic hypermutation (SHM) and class switch recombination (CSR) of antibody genes within the B-cell follicle of peripheral lymphoid organs. Ectopic overexpression of the enzyme leads to mutations in non-B cells and Escherichia coli (E.coli) genes. However, induction of mutations in E...

Origin, diversity, and maturation of human antiviral antibodies analyzed by high-throughput sequencing

Our understanding of how antibodies are generated and function could help develop effective vaccines and antibody-based therapeutics against viruses such as HIV-1, SARS coronavirus (SARS CoV), and Hendra and Nipah viruses (henipaviruses). Although broadly neutralizing antibodies (bnAbs) against the HIV-1 were observed in patients, elicitation of such bnAbs remains a major challenge when compare...
